Abstract [sv]

Artikeln bygger på en intervjuundersökning bland 35 av de 50 prefekterna vid Umeå universitet 2003-2004. Prefekter med kortare erfarenhet av prefektsysslan utelämnades. Syftet var att öka kunskaperna om prefekternas arbete med kvalitetsfrågor och få underlag för det universitetsgemensamma kvalitetsarbetet. Häften av prefekterna använder begreppet kvalitet återkommande i sitt arbete – övriga sällan eller aldrig.

Prefektintervjuerna avspeglar åtskilliga kvalitetsdefinitioner, strategier och åtgärder för att bedriva verksamheten med god kvalitet. Insatser inom grundutbildningen samt för att höja de anställdas kompetens är vanligast (2/3 resp. drygt 1/2 av fallen).

Kvalitetsåtgärder avseende forskarutbildning respektive infrastruktur nämns var dera i 1/5 av svaren. Forskning och

forskningsmiljö berörs bara undantagsvis.

Utvärderingar och uppföljningar (speciellt kursvärderingar) är vanligare kvalitetsåtgärder än utvecklingsarbete. Mindre än en femtedel av institutionerna bedöms ha en genomtänkt strategi för sitt kvalitetsarbete som man arbetar efter och kan peka på tydliga resultat. Drygt hälften av institutionerna beskriver ingen explicit strategi, det praktiska kvalitetsarbetet är begränsat till enstaka områden och resultaten är svåridentifierade. Resten (ungefär 1/3 av institutionerna) befinner sig däremellan.

Abstract [en]

The M-band is the transverse structure that cross-links the thick filaments in the center and provides a perfect alignment of the A-band in the activated sarcomere. The molecular composition of the M-bands in adult mouse skeletal muscle is fiber-type dependent. All M-bands in fast fibers contain M-protein while M-bands in slow fibers contain a significant proportion of the EH-myomesin isoform, previously detected only in embryonic heart muscle. This fiber-type specificity develops during the first postnatal weeks. However, the ratio between the amounts of myosin and of myomesin, taken as sum of both isoforms, remains nearly constant in all studied muscles. Ultrastructural analysis demonstrates that some of the soleus fibers show a diffuse appearance of the M-band, resembling the situation in the embryonic heart. A model is proposed to explain the functional consequence of differential M-band composition for the physiological and morphological properties of sarcomeres in different muscle types.

Abstract [en]

Type III-mediated translocation of Yop effectors is an essential virulence mechanism of pathogenic Yersinia. LcrV is the only protein secreted by the type III secretion system that induces protective immunity. LcrV also plays a significant role in the regulation of Yop expression and secretion. The role of LcrV in the virulence process has, however, remained elusive on account of its pleiotropic effects. Here, we show that anti-LcrV antibodies can block the delivery of Yop effectors into the target cell cytosol. This argues strongly for a critical role of LcrV in the Yop translocation process. Additional evidence supporting this role was obtained by genetic analysis. LcrV was found to be present on the bacterial surface before the establishment of bacteria target cell contact. These findings suggest that LcrV serves an important role in the initiation of the translocation process and provides one possible explanation for the mechanism of LcrV-induced protective immunity.
