Regulatory roles of small, nuclear-encoded proteins in the organisation of macro-complexes in photosynthetic membranes
Abstract [sv]
Photosystem II complexes, which are responsible for photosynthetically splitting water into oxygen, protons and electrons, are located mainly in the granal thylakoids of higher plants. Each complex consists of more than 30 protein subunits, with a total molecular mass exceeding 700 kDa. Eighteen of the subunits are low molecular mass proteins (< 10 kDa), all of which have a single transmembrane span, and their protein sequences are highly conserved among photosynthetic organisms. Today we have very little knowledge of their functions, or why so many of them are needed. Acquiring such knowledge could substantially improve our understanding of plants? photosynthetic performance and have potentially valuable applications also for other systems. Knock-out mutants that are deficient in one or other of the small proteins have been generated, but they have not as yet provided clear indications of their roles. However, we have recently obtained intriguing clues, since PsbW and PsbX knock-out plants show changes in phosphorylation patterns of PSII and the macro-structure of their thylakoid membranes. Aims: 1. Explore the mechanisms underlying the dramatic changes in PSII phosphorylation and the macro-organisation of grana thylakoid membranes following deletion of PsbW and PsbX proteins 2. Analyse in depth the roles of the two low molecular mass proteins PsbTn and PsbY within the PSII super-complex 3. Identify genes and proteins involved in PSII assembly and repair processes
