Our work focuses on structural and functional studies of RNAs and RNA-protein complexes (RNPs) based on X-ray crystallography. Flaviviruses are RNA viruses that cause serious diseases in humans. The protein coding region in the flaviviral RNA genome is flanked by 5´ and 3´untranslated regions which contain conserved RNA secondary structures. Some of these structures have been functionally characterized and shown to be essential for viral replication, translation and pathogenesis. The dengue virus serves us as a model system to investigate the structure-function relationships of these RNAs and their RNPs. We expect that these studies provide new insights into virus formation and pathogenesis and may lead to novel approaches of anti-dengue therapies. The Signal Recognition Particle (SRP) is a ubiquitous RNP which is essential for protein targeting in all cells. In the past decade a considerable amount of structural data from us and other groups have elucidated many details of the mechanistic basis of SRP function. However, still very little is known about the interaction with its most important ligand the signal sequence. We have crystallized SRP-signal sequence complexes and will complement the insights gained by the structural analysis with structure-guided mutagenesis to reveal the molecular determinants of signal sequence binding. These studies will be crucial for the understanding of this key-step in protein targeting and its regulation of the downstream events.
