Construction of optical tweezers instrumentation and Evaluation of its stiffness using the power spectrum method

Abstract [en]

Optical tweezers are powerful tools in biophysics, for example, optical tweezers can measure forces in pico-newton level, thus making it possible to quantify the mechanical properties of biopolymers. In this project, we realized an optical tweezers setup and evaluated the trap stiffness by the power spectrum method. Thus, in an experiment, the laser beam transmitted and scattered of a bead was collected and illuminated a photosensitive-detector. Using the power spectrum method and recording the Brownian motion of 1μm sized beads, we found that the trap stiffness of the optical tweezers was 3.4(3) pN/μm and 3.1(1) pN/μm in X- and Y-direction, respectively. Although we had problems in the beginning to realize the setup, we managed to trap several beads at the end.
