[{"_id":"project:1626","_type":"project","abstract":{"en":"Oxygen (O2) levels vary widely in the environment, which induces dramatic behavioral and physiological changes to resident animals. Adaptations to O2 variations can be either acute or chronic. Many fundamental aspects of acute and sustained O2 responses remain poorly understood at the molecular level. The research proposed here aims to explore the molecular and neural circuit underpinning of acute and chronic O2 sensing in the model system nematode C. elegans. We will combine high-throughput behavioral screen, biochemistry, calcium imaging, optogenetics and single neuron transcriptome profiling to systematically dissect O2 sensing in a whole animal system with a well-described nervous system. With many preliminary data already generated, it is very likely that substantial progression will be made within 2 years for acute hyperoxia (21% O2) and hypoxia (1% O2) sensing projects, and it is possible to identify a collection of molecules critical for anoxia (0% O2) tolerance in less than 2 years followed by functional dissection of these molecules. Research in C. elegans offers special advantages to address the intriguing and critical questions in O2 sensation, which are technically challenging in the other systems such as carotid body.
