Abstract [sv]
There is a need to develop rapid general and specific diagnosis of infectious disease as well as enable a rapid method (hours) for determination of antibiotic resistance patterns of bacterial pathogens. A most wanted need is to establish techniques that can be employed early in the healthcare system. We propose that global metabolomic profiling can be developed as a novel tool that would meet the above requirements. Our studies using plasma samples from infected mice, involving GC/MS and statistical multivariate analyses indicate that it metabolic profiling is a powerful method to detect even very mild infections and also distinguish between different bacterial pathogens. Specific infections could be detected in a background of other infections, which imply that the method will work also in human samples regardless of individual variation. Importantly, the method could be used to detect beta-lactamase-mediated bacterial lysis, implying it suitable for determination of antibiotic resistance. The aim of this application is to explore metabolic profiling as a diagnostic tool. We will define diagnostic biomarkers using the Yersinia mouse infection model, and investigate if metabolomics can be used for rapid and specific diagnosis of different types of infections and for detection of antibiotic resistance, first in the mouse and later in humans. This is a novel emerging technology that promises to be used to rapidly diagnose most infectious diseases from a simple blood sample
