Modeling the impact of macromolecular crowding and aggregation on biological processes

Recent years have witnessed a tremendous development in experimental techniques possesing unprecedented abilities to explore the micro- and nanoscopic world of cell biology (e.g. PALM and STORM). In order to fully exploit these achievements, quantitative modelling is crucial. I propose therefore two interrelated theoretical research projects:(i) Crowding effects on cellular processes. We will develop models for diffusion and binding of (fluorescently) tagged proteins (e.g. DNA transcription factors) when surrounded by other macromolecules. A proper microscopic description of such processes is a key to the understanding of regulatory and metabolic networks.(ii) Cellular response to unwanted protein aggregation. We will model the ongoing battle between aggregation growth and degradation by the cell´s protein quality control system. We quantify experimentally accessible observables such as timescales of in vivo growth and aggregation size distributions. An understanding of aggregation processes in cells is crucial in the fight against aggregation-driven neurological disorders (e.g. Parkinson´s disease).These projects will constitute the base from which I will kick-start my own research group dedicated to model biophysical problems. To boost scientific progress, world-leading theorists and experimentalists, with whom I have collaborated with during my postdoc time at the Niels Bohr Institute and New York University, will be brought into the proposed projects.
