After an acute infection, RNA viruses may remain in the human body as inapparent infections, potentially causing serious complications later in life. The mechanisms behind the establishment of such persistent infections are poorly understood.The fruit fly, Drosophila melanogaster, is now a well established model to study innate immune reactions and virulence mechanisms. My lab contributed to this development, identifying the first Drosophila genes of the innate defense. Recently we discovered a Drosophila RNA virus, the Nora virus, which causes persistent infections in flies. It is picorna-like, distantly related to e.g. the human poliovirus. I here propose to study the mechanisms of persistence in this virus, using advanced Drosophila and viral genetics to study persistence in large-scale experiments in vivo. This can not easily be realized with any other organism.We have generated an infective cDNA clone of the virus, allowing reverse genetics with the virus in vivo. I now propose to: 1. Study the effect of Drosophila mutations in selected genes. 2. Make microarray studies of gene expression in the infected flies. 3. Use RNAi to find host genes that affect the virus. 4. Make targeted mutations in the virus to identify viral persistence and virulence factors.A better knowledge of the basal mechanisms of viral persistence may lead to prevention of complications of RNA virus infections in humans, such as post-polio syndrome, chronic hepatitis and liver carcinoma.
