Our research addresses questions regarding mechanisms, expression and function, of genes and gene products in Escherichia coli and other enterobacteria that contribute to the bacterial interaction with host-cells. We study in particular different pathotypes of the species E. coli. This bacterium, which is part of the normal intestinal microbiota of humans, exhibits both physiological and metabolic versatility and includes, in addition to a variety of non-pathogenic, commensal variants, several pathogenic variants that can cause various types of intestinal or extraintestinal infections in man and animals. The studies will in particular focus on the following topics and questions: * Expression and role of the cytotoxic ClyA protein in commensal & pathogenic E. coli? * Is the clyA locus subject to epigenetic control? * Mechanism of translocation of ClyA protein to the bacterial periplasm? * The role of OMVs in release of bacterial protein toxins and interaction of OMVs with host cells and tissues: Can peptidoglycan intracellular delivery occur via OMVs? * Effect on bacterial surface and OMV properties by changes in LPS structure? * Function and role of newly discovered proteins from fimbrial gene clusters in ExPEC? * Monitoring adhesion and fimbrial biomechanical function by nanotechnology approaches
