PRESCRIBED DRUGS, GENES AND INJURIOUS FALLS IN MEN AND WOMEN

In a recent study we found that women have 80% more injurious falls than men and that this is the main reason for the higher fracture incidence in women than in men. The reason for the higher incidence of injurious falls in women is unknown. The overall aim of the present study is to quantify the genetic and environmental determinants of injurious falls in men and women. Environmental factors associated with injurious falls will be investigated in a cohort consisting of 40000 twins. Genetic factors associated with injurious falls will be investigated in a three cohorts including a total of 30000 subjects. Candidate genes include the estrogen receptor, SPTBN1, the vitamin D receptor and the cytochrome p450 system. In a final aim we will investigate whether there is a gene environment interaction between the use of antidepressants and the candidate genes with respect to injurious falls in the 30000 subjects above. The cytochrome p450 system is of special interest since antidepressants are metabolized by this system and a functional polymorphisms is known leading to a slow metabolism. Given the consequences of injurious falls, including pain, increased dependency and high costs for the health care system, increased knowledge about injurious falls would be important and could increase the possibilities for prevention. Identifying genes related to falling would also increase the knowledge about the genetic background to falling and could assist in the development of new drugs.
