Creation of Safe Antibacterial Surfaces- Antivirulence Gallium Complexes in Solution and at Surfaces
Abstract [sv]
Bacterial colonization of surfaces can create problems in many areas of our society, especially in health care with prolonged or reoccurring infections. The long term aim of this multidisciplinary project is to create antibacterial surfaces preventing this problem. The surfaces should also ideally be active against multi drug resistant bacteria. We will investigate Ga salicylidene acylhydrazide (hydrazone) complexes in solutions and in a polymeric surface film. Ga is a Fe antagonist and hydrazones have antivirulence effects on bacteria. In combination, the two display synergistic effects and the complex represents a new type of antibiotic with different mode of action compared to traditional antibiotics. At the end of this project we will understand the solution chemistry of the complexes, we will know their biological effects on Gram-negative and Gram-positive bacteria. We will know if the complexes are cytotoxic to mammalian cells and we will have optimised and tested antibacterial surface films incorporating the complexes. We will use chemical and microbiological analysis tools such as spectroscopy, thermodynamic modelling, chromatography, biological assays, flow cells and microscopy. The study will give us a good understanding of the mode of action and suitability of these new types of antibiotics for treatment of infections and in antibacterial surface films.
