Cells communicate with the outside world through receptors exposed on the cell surface. Receptors, which regulate nutrient uptake and senses the exterior, are constantly turned over to make sure that cells respond normally to surrounding stimuli. The systems that control uptake of receptors are built up of specific protein networks, which sculpture the membrane into vesicular carriers that bud and enter into the cell in a process called endocytosis. This process is fundamental to cells and malfunction results in sever human diseases such as cancer, muscle dystrophies and mental retardation. Furthermore, bacteria and viruses hijack the endocytic pathways in order to enter cells and establish infection.Our research aims to understand what triggers endocytosis and how endocytic carriers are generated. The focus of this project is to: (1) Screen for previously unknown components of endocytic machineries and characterise their specific action. (2) Study how cells take up viruses and bacterial toxins and identify receptors that are endocytosed via specific uptake carriers. (3) Establish biochemical in vitro reconstitution systems of carrier formation. (4) Develop advanced imaging methodology for ultrastructural determination. This knowledge can be used to specifically induce or block uptake, or improve specific drug-delivery and could provide solutions to the urgent need for novel effective drugs against infections and the demand for more stringent therapies against cancers.
