Abstract [sv]
The purpose of this project is to increase our understanding of how the nucleoid proteins in the H-NS protein family are influencing gene regulation in bacteria. In particular we aim to identify and characterize the mechanisms leading to differentiated sub-populations of bacteria. Specific goals: * To clarify the interplay between homologs/paralogues of the H-NS protein family involved in transcriptional silencing in extraintestinal E. coli isolates. * To clarify the molecular mode of action of regulatory proteins acting as anti-silencing factors alleviating H-NS activity in bacterial gene clusters encoding fimbrial adhesion organelles. * To identify and characterize the postulated regulasome complexes at the sub-cellular level in bacterial cells * To clarify the role and mode of action of bacterial nucleoid proteins in control of gene expression at post-transcriptional levels.We now find it feasible to study the actual mechanisms operating at the level of the particular sub-populations and the individual cells. Recent developments with fluorescence tagging approaches and super-resolution microscopy will allow us to identify and characterize both the spatial localization and composition of regulatory complexes in the individual bacterial cells.
