Chromosome-wide gene regulatory mechanisms

The only chromosome-wide targeting mechanisms recognised until recently were the dosage compensation mechanisms that equalize the transcriptional activity of the single male X-chromosome and the two female X-chromosomes. However, we have discovered the first known example of a chromosome-wide targeting mechanism for an autosome, dubbed Painting of Fourth (POF), in which POF and heterochromatin protein 1 (HP1) are involved in global regulation of the 4th chromosome in Drosophila melanogaster. POF binding is dependent on heterochromatin, and POF and HP1 bind interdependently to the 4th chromosome. We have proposed that a balancing mechanism involving POF and HP1 may provide a feed-back system that fine-tunes the expression status of genes on the 4th chromosome. An intriguing possibility is that such balancing mechanisms may regulate gene expression more generally at a chromosome-wide level. Our continuing analysis of chromosome-wide regulation is focused on four main issues: (1) The correct targeting of chromatin associated factors; (2) genome-wide fine-tuning of transcription output; (3) establishment and propagation of chromatin structure within and between chromosomes; and (4) the evolution and function of chromosome-wide targeting mechanisms. We believe that our work has the potential to make major contributions to our understanding of chromosome-wide regulatory complexes, their composition, function and evolution.
