Abstract [sv]
By further developments and refinements of our metabolomics and chemometric bioinformatics approach we aim to increase the information output from metabolomics data. We will use multivariate methods for extraction of ´latent biomarkers´ as for allowing robust and high-sensitivity analyses in large scale metabolomics studies. In addition, we will apply and develop strategies for using metabolomics data as intermediate phenotypes in associations to alterations in the genome (or other `omes´). Finally and importantly, the methods will be applied to important areas in cancer (prostate, brain, pancreas), with the aims to find biomarkers for, i) early diagnosis, prognosis and therapy prediction, ii) tumour progression to reveal novel therapies and iii) sensitive detection of disease sub groups to aid individualized treatment. This will potentially lead to a more sensitive diagnostics of different cancer types at a curable time point and importantly stratification of tumour aggressiveness at diagnosis. Another intriguing prospect is monitoring of treatment effects in close to real time to allow individualized treatment. A more sensitive definition of disease sub groups is particularly interesting in combination with alterations in the genome where the metabolomics data could act as intermediate phenotypes and help identifying risk genotypes and link to altered pathways. Such an approach could reveal new therapeutic options and facilitate the development of individualized therapies.
