AMPK ACTIVATOR O304: A POTENTIAL CALORIC RESTRICTION MIMETIC DRUG FOR AGE-RELATED DISEASES?

With the increasingly ageing population, chronic, age-related diseases like diabetes, cardiovascular disease, cancer, and neurodegenerative diseases become more prevalent. Caloric restriction (CR) is one of the few regimens with documented beneficial anti-ageing health effects in a huge variety of tested model organisms. Thus, drugs that mimic the effects of CR has gained attention as an attractive and feasible strategy for age-related diseases. Activators of AMP activated protein kinase (AMPK) represents one such class of potential geroprotector drugs. We have recently identified a novel PAN-AMPK activator, O304, which mimics the beneficial metabolic and cardiovascular effects of caloric restriction in diabetic and obese mice and in type 2 diabetics on Metformin. Thus, this proposal aims at elucidating the metabolic, cardiovascular, physical, tumourogenic, and cognitive effects of O304 in  ageing mice. Metabolic, cardiovascular, physical, and cognitive tests will be performed at start and during ageing. Necropsies and isolation of tissues for further analyses will be done after sacrifice or death. Given that O304 was well tolerated with no adverse effects in pre-clinical, Phase I and Phase IIa clinical studies, and proved to have beneficial metabolic and cardiovascular effects in a Proof-of-Concept Phase IIa study in T2D diabetics on Metformin, O304 thus holds great potential as a drug that can boost resilience to multiple age-related diseases in humans.
