The project aims at providing a tool ? PREDATAR - to increase the usability of in silico and in vitro data in chemical risk assessments, which is a key in the process of replacing, reducing, and refining animal testing. A cornerstone is estimation of dose at target using physiologically based toxicokinetics (PBTK) modeling. Non-testing methods including in silico and in vitro methods typically neglect administration, distribution, metabolism and excretion processes and may provide high ratios of false positives and negatives. These weaknesses could be solved by the tool and thus increase the use of alternative testing methods. We intend to apply the zebrafish embryo toxicity test for measurement of toxicological key events, which provides a means to test chemicals in an intact organism defined as an alternative method to animal testing. A novel PBTK model for zebrafish embryos will be developed that would enable improved understanding of effects measured in the assay and increased use in risk assessments. PREDATAR will be developed and validated on the adverse outcome pathways of agonistic and antagonistic estrogen receptor activity leading to reproductive disorders. Reproductive disorders can be identified by several protocols using large numbers of animal tests. The project will provide a means for substantial reduction of animal testing and a break-through in the intercept of the emerging field of computational exposure sciences and pathway based risk assessment.
