Abdominal aortic aneurysm (AAA), is an enormous health care burden and ruptured AAA account for approximately 200,000 of annual deaths world-wide. AAA is an inflammatory disease and despite anti-inflammatory treatment due to other underlying vascular disease these patients have, these drugs neither prevent nor inhibit aneurysm disease. Today, no treatment exists for AAA. To identify therapeutic strategies to prevent or stabilize AAA, the molecular mechanisms behind disease progression need to be elucidated.Aneurysms develop as a consequence of weakening of the vascular wall associated with perivascular inflammation. By combining patient samples, mouse models of AAA and reductionistic cellular approaches, my laboratory has identified several key factors in the development of AAA that we believe can be targeted to inhibit disease development. The main aim with the current proposal is to further clarify pathophysiological and molecular mechanisms behind AAA disease and to test novel treatment strategies. Using our translational approach, where we investigate effects on immune cell activation and vascular remodelling by transcriptional regulation and by targeting glycolysis and kinase activity of certain proteins using available drugs.To identify therapeutic options to retard or prevent development of AAA is of outermost importance, and I believe that a translational approach is necessary to uncover mechanisms driving the disease for future drug development.
