Endocytic membrane remodelling machineries and their impact on cell adhesion

Endocytosis is the way by which cells take up nutrients, regulate cell surface receptors and control membrane rearrangements during migration. The research aims for a thorough understanding of proteins and mechanisms that are used by cells to remodel membranes into carriers for endocytic transport. The project focuses on trafficking driven by the membrane remodelling proteins GRAF1 and EHD2 which mark distinct routes into the cell. Common for these pathways, is that they the facilitate uptake of molecules that are clustered in lipid-dependent microdomains of the plasma membrane. Our previous findings show that these pathways also are important for the morhology and behaviour of cells. We will characterise how these pathways are used to regulate cell adhesion and migration, with the specific aims to:I. Understand the role of GRAF1-dependent endocytosis during down-regulation of focal adhesions and regulation of cell motility. II. Define the cellular role of EHD2 in structure-control and clustering of cell surface caveolae.
