BACKGROUND, SIGNIFICANCE. Our innate immune defenses are still incompletely understood. To address this problem, we have developed Drosophila as an experimental model. The usefulness of this model was demonstrated for instance by our discovery that the receptor Toll can activate an antibacterial response in Drosophila. Toll-like receptors were later found to play an important role in human immunity. In fact, the entire signaling pathway is conserved between insect and man.AIMS. To explore the basic molecular mechanisms of innate immunity, taking advantage of the possibilities offered by Drosophila as a model system. Specifically, we will focus on cellular immunity and on the viral defense.WORKING PLAN. We have now developed novel tools to study the activation of Drosophila blood cells in response to infection. Using genetic screens we will characterize the genes involved in this response. In a gain-of-function screen we recently identified several signaling pathways that are involved. In this project, we will search for relevant genes in modifier screens, using deletions that cover at least 85% of the genome. The biological role of specific genes will be studied in the immune response against the parasitoid, Leptopilina boulardi. Furthermore, we will use an RNA virus that we recently discovered to study the genetics of Drosophila/virus interactions, and to find out how persistent viral infections are formed.
