The invasive tumor front in colorectal cancer
Abstract [sv]
The clinical behaviour of a colorectal cancer (CRC) is very much reflected on the course of events at the invasive front. We have collected tissue from 800 CRC patients. In clinical materials, we want to: A) investigate the clinical role of EMT (epithelial-mesenchymal transition) that takes place at the invasive front. The mechanisms involved will be studied in a novel three-dimensional invasion model in vitro including co-culturing of different cell types in an attempt to imitate the true microenvironment. B) Using the same design, the ?migrating cancer stem cell concept? will be further evaluated. C) The role of macrophages in the front will be studied both in the tissue material and in experimental models. Are different types of TAMs present at the front? M1 or M2 types? In that case, why are different subtypes recruited? D) We want to study the effects of tumor-associated fibroblasts and their contribution to invasion and metastasis. Herein we present very interesting preliminary results when studying the fibroblast activation protein (FAP). E) A promising novel classification of CRCs are based on the CIMP status (CpG Methylator Phenotype)(CIMP negative, 1,2). CIMP status will be analysed by MethyLight technique in the tumor tissue samples and the data will be correlated to the setup above incl. survival. In conclusion, better understanding of the molecular events at the invasive front will be crucial to design better future management for colorectal cancer patients.
